Stanley made the decision to play college football for the University of Notre Dame in December 2011. He was a player for the Notre Dame Fighting Irish. Stanley competed in two games as a reserve as a true freshman in 2012. As a sophomore right tackle in 2013, he started all 13 of the games.
He participated in all 13 games during his junior year in college. After his senior year, he made it known that he intended to register for the 2016 NFL Draft. He was chosen by the Baltimore Ravens in the first round of the 2016 NFL Draft. He received a four-year, $20.48 million rookie contract with the Baltimore Ravens in May 2016 along with a $13.09 million signing bonus.
He made 15 starts in 2018 and only missed one due to an ankle ailment. The Ravens chose to exercise Stanley’s fifth-year option in April 2019. He and the Ravens agreed to a five-year, $112.8 million deal in October 2020. In October 2021, he underwent season-ending ankle surgery.
