Madubuike was born in Dallas, Texas, to parents who were originally from Nigeria. Madubuike was a student at Texas McKinney North High School.
He was chosen to participate in the 2016 All-American Game that was held by the United States Army. He made a verbal commitment to play collegiate football for Texas A&M University.
Madubuike redshirted during his freshman season at Texas A&M in 2016. He participated in all 13 games that were played in 2017, accumulating 20 tackles. In 2018, he played in all 13 games once more and finished the season with 40 tackles and 5.5 sacks.
As a junior, he recorded 45 tackles and 5.5 sacks on the football field. After the season, he made the decision to forego playing in his senior year and instead join the NFL Draft in 2020. He made the decision to not participate in the 2019 Texas Bowl.
In the third round of the 2020 NFL Draft, the Baltimore Ravens selected Madubuike with the 71st overall pick, making him the team’s third overall selection.
On November 27, 2020, the team put him on the reserve/COVID-19 list. He was brought back onto the active roster on December 7, 2020.
