Holmes started playing football at a young age. He attended McNair High School in Atlanta, Georgia, and played high school football. As a utility player, he rotated between being a cornerback and wide receiver.

Holmes initially attended the University of Missouri and played four seasons with the Missouri Tigers. He played sparingly in 10 games as a true freshman for the Missouri Tigers, making just one tackle.
Because he hurt his shoulder in a preseason game, he would have to sit out his second year. As a redshirt sophomore, Holmes started eight of the 13 games he appeared in and had 12 pass breakups.
Holmes transferred to Oklahoma State University and played for the Oklahoma State Cowboys from 2020 to 2021.
