Moore was enlisted to the Rays on September 11, 2011, and made his major league debut three days later, surrendering a two-run home run to Matt Wieters in the eighth inning of a 6–2 loss to the Baltimore Orioles.

On September 22, he made his maiden major league start, pitching 11 strikeouts in five innings or less against the New York Yankees, becoming the first pitcher in MLB history to do so.

The Rays won 15–8. Moore started Game 1 of the 2011 American League Division Series (ALDS) against the Texas Rangers, pitching seven scoreless innings and leading the Rays to a 9–0 victory

Moore had a 1–0 record and a 2.89 ERA in 9+13 innings in his debut major league season.

Moore signed a guaranteed five-year, $14 million contract with the Rays on December 9, 2011.

His contract was the greatest ever in terms of guaranteed money and prospective earnings for any pitcher with less than two years of service time, and it was part of a Rays organization-wide pattern of awarding long-term contracts to young pitchers.

Moore joined the Rays’ starting rotation in 2012 as part of a five-man group that includes James Shields, David Price, Jeremy Hellickson, and Jeff Niemann.

Moore and relievers Burke Badenhop and Brandon Gomes combined to pitch a one-hit shutout against the Miami Marlins on June 15, winning 11–0 and snapping the team’s three-game losing run.

After going 5–1 with a 3.94 ERA in eight starts in June and July, the Rays gave away a Moore bobblehead figure during their game against the Seattle Mariners on July 22.

Moore had an 11–11 record, a 3.81 ERA, and 175 strikeouts in 31 games over 177+13 innings this season.
