In 2018, he participated in all 14 games and started 12 of them. In 2019 and 2020, he started at center for Oklahoma and was recognized as the best offensive lineman in the Big 12 Conference in both of those years.

Humphrey made the announcement following the conclusion of the 2020 season that he would not be returning to school for his last year of eligibility and instead enter the 2021 NFL Draft.
