As an undrafted free agent, Bourne was signed by the San Francisco 49ers to a three-year, $1.67 million deal on April 30, 2017. A $25,000 guarantee and a $5,000 signing bonus were part of the agreement.

He played in his first regular-season game as a professional during the San Francisco 49ers’ 23-3 loss to the Carolina Panthers in the season opener.

For the following five games, Bourne did not participate as a healthy scratch (Weeks 2–6). On November 5, 2017, Bourne hauled in two catches for 39 yards while the Arizona Cardinals were defeated 20-10.

Bourne agreed to a three-year, $15 million deal with the New England Patriots on March 19, 2021. When the Patriots fell 35-29 in overtime in Week 6 to the Dallas Cowboys, Bourne produced a 75-yard touchdown on his sole catch of the game.

Nelson Agholor caught a 25-yard touchdown throw from Bourne in Week 7 against the New York Jets, marking the quarterback’s first career score.

Bourne contributed to the Patriots’ 36-13 victory over the Tennessee Titans in Week 12 with 5 receptions for 61 yards and 2 touchdowns, including one of 41 yards.
