The Philadelphia Eagles selected Gainwell in the fifth round of the 2021 NFL Draft, 150th overall. Gainwell added that Sproles was the inspiration for his style of play. On June 3, 2021, he agreed to a four-year rookie deal with Philadelphia.

Gainwell had a passable first season, finishing with 33 receptions for 253 yards and one score in addition to 68 carries for 291 yards and five touchdowns.

Gainwell played in 16 games as a rookie after sitting out the Eagles’ game against the New Orleans Saints in week 11.
