He played collegiate football for the Alabama Crimson Tide while attending the University of Alabama.

In 2017, he redshirted his true freshman year and participated in all 15 of the Crimson Tide’s contests. He made the Southeastern Conference (SEC) All-Freshman team after playing in 15 games and recording 18 tackles and a fumble recovery.

In the second round, the Washington Commanders selected Mathis with the 47th overall pick after he declared his intention to enter the 2022 NFL Draft following the regular season. On June 21, 2022, he agreed to a rookie deal for four years.
