When he was a student at the University of Alabama, he participated in NCAA football for the Alabama Crimson Tide. He redshirted his true freshman year of college in 2017 and took part in all 15 of the Crimson Tide’s games. After participating in 15 games, registering 18 tackles, and recovering a fumble, he was named to the Southeastern Conference (SEC) All-Freshman team.

He played in 12 games in 2019 and finished with 27 tackles and 0.5 stops for loss. Mathis announced his intent to enter the 2022 NFL Draft following the regular season, and the Washington Commanders picked him with the 47th overall pick in the second round. He committed to a four-year rookie contract on June 21, 2022.
