Stanley made his college football commitment to the University of Notre Dame in December 2011. He participated in football for the Notre Dame Fighting Irish. He started each of the 13 games he played in as a junior in 2014, surrendering only one sack all season. He made an official announcement to enter the 2016 NFL Draft following his senior year. He was chosen by the Baltimore Ravens in the first round of the 2016 NFL Draft. He was given a four-year, $20.48 million rookie deal by the Baltimore Ravens in May 2016, along with a $13.09 million signing bonus.

He made 15 starts in 2018 and missed one due to an ankle ailment. The Ravens exercised the fifth-year option on Stanley’s contract in April 2019. He signed a five-year, $112.8 million agreement with the Ravens in October 2020. After undergoing season-ending ankle surgery in October 2021, he was put on injured reserve.
