The Colorado Rockies selected him in the first round of the 2018 MLB draft. He signed with the Rockies and was given $2.9 million as a signing bonus. He played for the Grand Junction Rockies of the Rookie-level Pioneer League that season to kick start his professional career.

He spent the following season with the Asheville Tourists of the Class A South Atlantic League. That same season, the Rockies promoted him to the Lancaster JetHawks of the Class A-Advanced California League. When the season resumed in 2021, he played for the Hartford Yard Goats of the Double-A Northeast. He was later promoted to spend the rest of the season with the Albuquerque Isotopes of the Triple-A West league. In June 2021, he was placed on the injured list after having his appendix removed.

The Rockies added him to their 40-man roster in November 2021. In June 2022, the Rockies announced that Ryan Rolison was scheduled for surgery and is expected to miss the entire 2022 season.
