Mayza pitched for the Class-A Lansing Lugnuts throughout the 2015 season. He pitched in 26 games for the Lugnuts, posting a 3–2 record, 3.07 ERA, and 62 strikeouts in 552/3 innings.

Mayza advanced through the Blue Jays system, starting the 2016 season with the Advanced-A Dunedin Blue Jays.

He was promoted to the Double-A New Hampshire Fisher Cats in June and spent the rest of the season with them.

Mayza appeared in a career-high 42 games in 2016, going 3–3 with a 2.25 ERA and 65 strikeouts in 64 innings. Mayza was invited to MLB spring training on January 19, 2017.

To begin the 2017 season, he was assigned to Double-A New Hampshire. Mayza was promoted to the Triple-A Buffalo Bisons after 29 appearances.

Before his first promotion to the Majors, he made 11 bullpen appearances with a 0.93 ERA.

 

Mayza was called up by the Blue Jays on August 14, 2017.

On August 15, he made his major league debut, pitching a scoreless ninth inning in a 6–4 loss to the Tampa Bay Rays and striking out Peter Bourjos for the first time.

On September 12, Mayza earned his first major league win, pitching the bottom half of the ninth inning in a 3–2 Blue Jays victory over the Baltimore Orioles.
