Marcano did not play in a game in 2020 since the minor league season was canceled due to the COVID-19 epidemic. After the 2020 season, the Padres added him to their 40-man roster.

Marcano made his Major League Baseball debut on April 1, 2021, as a pinch hitter for Keone Kela, drawing a walk against Arizona Diamondbacks pitcher Stefan Crichton.
