Being an offensive tackle with the Arizona Cardinals of the National Football League, D. J. Humphries is a well-known professional football player. At 1.96 m tall and 139 kg, he is both tall and heavy. When he was a student at Mallard Creek High School, he played three sports: football, basketball, and track. He participated in Florida Gators football games from 2012 to 2014 for the University of Florida.

As a true freshman, he played in all 12 of the games, starting three of them. He was awarded the Sporting News Freshman All-America and first team Offense SEC Coaches’ All-Freshman (OL) accolades. He participated in the first seven games of his second season before suffering a season-ending injury in 2013.

He played in 10 games as a junior during the 2014 season. After his junior year, he declared for the 2015 NFL Draft. The Arizona Cardinals selected Humphries with the 24th overall choice in the 2015 NFL Draft. In that year, he signed a $4.74 million signing bonus and a four-year, $8.91 million rookie contract with $7.26 million guaranteed.

In April 2018, the Cardinals picked up the fifth-year option on his contract. In February 2020, he agreed to a three-year, $45 million contract extension that would keep him with the Cardinals through the 2022 season. On August 2, 2022, he agreed to an extension with the Cardinals that would pay him $66.6 million over three further years.
