Castillo became an international free agent in March 2014 and signed with the Tampa Bay Rays.

That season, he made his professional debut with the Dominican Summer League Rays, where he went 3–3 with a 3.96 ERA in 25 innings.



In 18 bullpen appearances with the Hudson Valley Renegades and the Bowling Green Hot Rods in 2015, he went 12–2 with a 3.03 ERA.

Castillo was a member of the Montgomery Biscuits and Durham Bulls in 2017. He was named the Rays’ minor league reliever of the year after pitching 71.2 innings and compiling a 2.76 ERA with 90 strikeouts and 15 saves.

After the season, the Rays added him to their 40-man roster. The Durham Bulls were his first team in 2018.

On June 6, 2018, Castillo was promoted to the Tampa Bay Rays and made his debut.   He spent the season pitching out of the bullpen and as one of the Rays’ “opening” starters.

Castillo was 4–2 in 11 starts and threw 56+23 innings in 43 games. He had a 3.18 ERA, 65 strikeouts, and 18 walks throughout the course of the season.

Castillo was utilized as a closer and opener in 2019. He was placed on the 10-day injured list with shoulder inflation on June 23.

He appeared in 65 games throughout the season (6 starts). He had 81 strikeouts, 8 saves, and a 3.41 ERA in 68+23 innings. Castillo pitched two shutout innings in the American League Wild Card game against the Oakland Athletics
