Desjarlais was selected by the Winnipeg Blue Bombers in the first round of the 2019 CFL Draft as the fourth overall pick, and on May 11, 2019, he signed a contract with the organization despite garnering attention from the NFL.

Following training camp, he was added to the team’s active roster, and on June 15, 2019, against the BC Lions, he made his professional debut.

Desjarlais skipped the 2020 CFL season as it was postponed. In a condensed 2021 season, he participated in all 14 games in the regular season.

At the conclusion of the campaign, Desjarlais was selected as a West Division All-Star. He started both of the postseason games as the Blue Bombers defeated the Tiger-Cats to win the 108th Grey Cup for a repeat victory.

Desjarlais was released in order to pursue a deal with the National Football League as a potential free agent.

Desjarlais and the New England Patriots agreed to a reserve/future deal on January 27, 2022.
