In January 2011, Aquino joined the Cincinnati Reds as an international free agent from the Dominican Prospect League.
He made his professional debut with the Dominican Summer League Reds that same year, and he also spent 2012 with them.
In 2013, he played for the Arizona League Reds and the Billings Mustangs; in 2014, he played for the Billings Mustangs; and in 2015, he played for the Billings Mustangs and the Dayton Dragons.
He was named the Florida State League Player of the Year in 2016 while playing for the Daytona Tortugas. After the season, the Reds added him to their 40-man roster.

On November 30, 2018, he was non-tendered and became a free agent. On December 3, 2018, he re-signed a minor league contract.
The Louisville Bats was his first stop in the 2019 season. The Reds chose Aquino’s contract on August 1. Aquino tied Trevor Story’s record of seven home runs in his first ten major league games.
Aquino hit three home runs on August 10, 2019, against the Chicago Cubs, becoming the first rookie in MLB history to hit a home run in three consecutive innings and the second rookie after Bobby Estalella to have a three-homer game in his first ten career games.
