The Cubs promoted Bryant to the majors on April 17, 2015. When he made his Wrigley Field debut that day, he went 0-for-4 with a trio of strikeouts. Bryant’s first at-bat came the next day at Wrigley Field when he hit an RBI single.

It was on May 9 of 2015  that Bryant hit his first major league home run. Two days later, against the New York Mets and Jacob deGrom, he hit his second home run at Wrigley Field.

It was Bryant’s best month of the year so far with seven home runs and 22 RBIs to go along with his.265 batting average and 16 walks. He was selected NL Rookie of the Month for May.

Bryant’s first career grand slam came in the ninth inning of a 17–0 triumph over the Cleveland Indians on June 17 against David Murphy. Bryant hit a two-run home run and a grand slam off Jarred Cosart on July 4 against the Miami Marlins.

He became only the second Cubs rookie since Billy Williams in 1961 to hit two grand slams. Giancarlo Stanton was forced to withdraw from the All-Star Game due to injury, and Bryant was named as his replacement on the National League’s roster in Cincinnati, Ohio.

He also competed in the Home Run Derby at the tournament. A no-hitter by Cole Hamels was completed on July 25 when DeMarcus Bryant struck out at the warning track for the last out.

On July 27, Bryant blasted a two-run walk-off home run off John Axford in a 9–8 victory over the Colorado Rockies, his first of his career.
