On January 6, 2016, Rondón signed as an international free agent with the St. Louis Cardinals.

A rookie in the Dominican Summer League, he went 2–2 with a 2.79 earned run average in 42 innings for the Rookie-level Cardinals that year.



Dominican League Cardinals, Gulf Coast Cardinals, and Johnson City Appalachian League a combined 3–4 with a 3.14 earned run average in 14 games amongst the three organizations in 2017.

During his time with the State College Spikes of the Class A Short Season New York–Penn League and the Peoria Chiefs of the Class A Midwest League in 2018.

He pitched to a 3–6 record and a 3.17 ERA in 15 starts, striking out 80 batters over 88 innings with both teams, and in 2019, he pitched to an 8–0 record and an ERA of 2.96 in two starts with the Palm Beach Cardinals of Class A-Advanced Florida State League (where The Cardinals selected Rondón their Minor League Pitcher of the Year after the season.

The COVID-19 outbreak forced the termination of the minor league season in 2020, hence Rondón did not play in any minor league games.

The Cardinals will have him on their 40-man roster after the 2020 season. He was assigned to the Memphis Redbirds of the Triple-A East League for the start of the 2021 season.

Rondón made his main league debut on June 5, 2021, when he was promoted from the minors for the first time.

The following day, he made his Major League Baseball debut against the Cincinnati Reds, tossing a scoreless inning in relief. Against Jonathan India, he recorded his first major league strikeout.
