In the television series Magnum, P.I., which was shown on CBS from 1980 through 1988, Mosley played the part of Theodore “T.C.” Calvin, a helicopter pilot.

This role brought Mosley the most fame. Mosley was married to Antoinette Laudermick, with whom he had been involved in a relationship for about sixty years prior to their marriage.

On August 4, 2022, he was engaged in an automobile accident, and he passed away on August 7 of the same year.

In the biographical film about the artist Leadbelly that was released in 1976 and directed by Gordon Parks, Mosley portrayed the titular character.

Terminal Island featured him in the role of Monk (1973). Tom Selleck, Phyllis Davis, Don Marshall, and Ena Hartman were some of the other performers that appeared in the feature film.

On March 11, 2019, Mosley made a surprise return to acting with a guest spot on the Magnum, P.I. reboot episode “A Kiss Before Dying,” playing the role of Booky, T.C.’s barber.
